| TLR4 signaling to both the MyD88 pathway and the TRIF pathway | |

| TLR4 associates directly with two TIR domain-containing adaptor molecules, TIRAP, which associates with MyD88, and TRAM, which associates with TRIF. In all cases, the TIR domains of these proteins mediate the associations. Thus, TLR4 signaling induces the transcription of both proinflammatory cytokines and interferon-β. TIRAP and TRAM are not known to connect to any additional signaling events beyond the MyD88 and TRIF signaling pathways. |
